Resumo

A região amazônica é rica em biodiversidade com elevado potencial econômico em diversos setores dentre os quais a pouca explorada área farmacêutica. De fato, somente 5% dos fármacos disponíveis no mercado de medicamentos são produtos naturais na sua forma original como foi extraída da natureza, 55% destes insumos farmacêuticos ativos (IFAs) passaram por modificação molecular ou serviram como modelos para a criação de novos medicamentos e os 40% restantes são de origem sintética. Esta região tem poucos profissionais e nenhuma empresa para desenvolver este setor. Assim, este projeto visa aplicar métodos de farmacologia e toxicologia celular e molecular para a seleção de compostos naturais e sintéticos bioativos usando moléculas isoladas da região amazônica, modificadas e sintetizadas com o auxílio de métodos de síntese orgânica e química medicinal. O estudo de farmacologia e toxicologia molecular inicia pela seleção de moléculas por simulação computacional e as modificações propostas por modelagem molecular com base em seus mecanismos de ação. As moléculas selecionadas serão sintetizadas por métodos clássicos para serem avaliadas por métodos enzimáticos e cultura de células. Os derivados avaliados visam atender a demandas de novos IFAs com alto valor agregado a fim de atender ao sistema único de saúde (SUS) e as indústrias farmacêutica, cosmética e nutracêutica. Abstract: The Amazon region is rich in biodiversity with high economic potential in various sectors, even their pharmaceutical area has been explored few. However, only 5% of the drugs available on the market are natural products in their original form extracted from nature; 55% of these active pharmaceutical ingredients (APIs) have undergone molecular modification or served as template for the creation of new drugs; and the remaining 40% are of synthetic origin. The region has few professionals and no companies to develop this sector. This project aims to apply methods of pharmacology and toxicology molecular and cellular to select bioactive natural and synthetic compounds using molecules isolated from the Amazon region, modified, and synthesized with the aid of organic synthesis and medicinal chemistry methods. The study of pharmacology and molecular toxicology begins with the selection of molecules through computational simulation and the proposed modifications through molecular modeling. These candidate molecules will be synthesized by classical methods to be evaluated by enzymatic and cell culture methods. The evaluated derivatives aim to meet the demands for new APIs with high added value to serve the Brazilian Unified Health System (SUS) and the pharmaceutical, cosmetic, and nutraceutical industries.
